      "description_narrative":["Illegal wildlife trade (IWT) is a widespread and lucrative criminal activity causing major global environmental and social harm. The IWT has been estimated to be worth up to £17 billion a year. Nearly 6,000 different species of fauna and flora are impacted, with almost every country in the world playing a role in the illicit trade. The UK government is committed to tackling illegal trade of wildlife products and is a long-standing leader in efforts to eradicate the IWT. Defra manages the Illegal Wildlife Trade Challenge Fund, which is a competitive grants scheme with the objective of tackling IWT and, in doing so, contributing to sustainable development in developing countries. Projects funded under the Illegal Wildlife Trade Challenge Fund address one, or more, of the following themes: • Developing sustainable livelihoods to benefit people directly affected by IWT, • Strengthening law enforcement, • Ensuring effective legal frameworks, • Reducing demand for IWT products.       "description_narrative":["In the last 15 years 260 million clinical cases of malaria have been prevented in Sub Saharan Africa, overwhelmingly through the use of insecticides that target the mosquito vector. Insecticide resistance is therefore a major threat to the sustained control of malaria. Whole genome sequencing of malaria mosquitoes has the power to revolutionise our understanding of the evolution of insecticide resistance; allowing us to design new insecticides and to develop strategies that delay the onset of resistance. However genomic technologies will only reach their full potential with improved sampling design and when extensive individual and population level metadata (collection location, infection/ disease state etc) accompany each sequenced sample. For the problem of insecticide resistance in the major malaria vector Anopheles gambiae a deep understanding of the biology lies with SSA scientists whilst the skills to sequence and analyse the data are held by centres in the North. This foundation proposal is designed to draw together these two groups with support from a world-leading spatial statistics group around the key issue of insecticide resistance evolution. Our vision is to further develop the programme to a Pan-African level, eventually devolving leadership to our SSA partners who will have the skills and experience to sustain the network.","The Global Challenges Research Fund (GCRF) supports cutting-edge research to address challenges faced by developing countries.       "description_narrative":["Malaria infects over 200 million people every, mostly in sub-Saharan Africa. Nearly half a million people died from the disease in 2015, the majority of deaths being in children under the age of five. The malaria parasite is spread by infected mosquitoes and the most effective way to monitor the disease is to monitor populations of these mosquitoes. However, current tools for sampling malarial mosquitoes are time-consuming and labour intensive, making them expensive and difficult to standardize. Accordingly, data between countries and regions cannot be reliably compared. To solve this problem, we have developed a mosquito trap that exploits the blood-seeking behaviour of mosquitoes by mimicking the sensory stimuli that a mosquito follows when searching for a person to bite. These include the look, smell and temperature of warm-blooded hosts. We have incorporated these stimuli into a trap that lures mosquitoes towards it and then captures them when they land. Our project will test this \"Human Decoy\" Trap against current methods used in mosquito monitoring to determine whether the Human Decoy Trap can overcome the limitations of existing tools. We will evaluate if Human Decoy Trap catches are suitably similar to those of current traps in terms of numbers of mosquitoes and other important data related to malaria that can be extracted from mosquitoes caught by traps, such as the proportion of caught mosquitoes infected with malaria parasites. Over 90 countries, mostly low and middle income, are malaria-endemic. We will work in Burkina Faso, Benin and Cameroon, three West African countries where malaria causes thousands of deaths every year, but differing in intensity and seasonality of transmission and with different mosquito species involved. This will help us to understand whether there are differences in trap performance in a wide range of malaria settings. In addition, we will work with end-users of the trap, namely local communities, public health operatives and field technicians, to better understand their perspectives and needs regarding mosquito sampling and control. During the course of the project, we will use data we collect to optimize and improve the trap's performance and design. This will help us to develop a commercial prototype that is effective and acceptable to end-users, maximising the likelihood the trap will be adopted into the communities and sectors that need it the most. Longer-term, we envision a version of the Human Decoy Trap that can be deployed as a mosquito control tool. The most effective way of controlling malaria is to reduce the number of infective mosquito bites a person receives. This is currently achieved by providing people with insecticide-treated bed nets to protect them from bites whilst they sleep and spraying the walls inside houses thus killing mosquitoes that rest there. However, neither of these options protects people from mosquitoes that may bite them outdoors during the day or just before they go to bed at night. Data from this current project will be used to provide preliminary evidence for the suitability of the Human Decoy Trap as a control tool that is specifically targeted at malaria mosquitoes biting outdoors, which may also be effective against other species of mosquitoes that can carry other infectious diseases, such as dengue fever, chikungunya and Zika viruses. Such an improved sampling method would enhance the quality of data necessary for efficient targeting and evaluation of malaria control intervention activities and reduce the cost of collecting it. A successful outdoor mosquito control device would help to reduce the population of infected mosquitoes that cause malaria.       "description_narrative":["Neglected tropical diseases are a diverse group of diseases that thrive mainly among the poorest of the world's populations. Three of the World Health Organisation's 10 most significant neglected tropical diseases - African trypanosomiasis (also known as sleeping sickness), leishmaniasis, and Chagas' disease - are caused by closely-related single-celled parasites. Human infection with African trypanosomes is brought about when an infected tsetse fly takes a blood meal. Without treatment the disease progresses through general ill health to coma and death. Current drugs in use against trypanosomes are old, toxic and failing due to emergence of resistance. Urgent new research is needed to identify potential new therapeutic options.  An unusual aspect of African trypanosomes is that they multiply in the human blood in full view of the body's defence systems. They do this by periodically changing their cell surface to escape recognition by the host. However, many molecules on the parasite surface perform essential functions and cannot be changed. Trypanosomes place these in a special, protected domain on the cell surface. Identifying surface-exposed invariant molecules and understanding how this protective segregation is maintained are of major scientific interest, as well as of practical utility in uncovering ways in which trypanosomes may be vulnerable to new therapies.  Using a combinatorial approach funded by the MRC, I have previously identified the composition of the African trypanosome cell surface. I now aim to exploit this knowledge to single out those invariant surface molecules that are essential to the survival of the parasite during infection. For this, I propose to harness some of the power of modern DNA sequencing technologies to test which cell surface genes, when silenced, cause the parasite to die inside the host. A similar method will be used to identify not only cell surface genes, but any genes that the parasite uses to maintain the cell surface organisation, which is so critical to escaping the host immune attack.   Finally, I propose to test those surface-exposed molecules for their potential as vaccines. My pilot experiments show that I can use genetically-modified parasite to screen for molecules on the parasite surface that are accessible to the immune system. I propose to develop this assay and test the most promising candidates in an animal model of human disease.       "description_narrative":["Tuberculosis (TB) is a major cause of disease and death globally. In 2015, WHO estimated there were 9.6 million TB cases and 1.5 TB deaths. Nearly 500,000 of these cases were resistant to two or more of the main drugs used to treat TB. New drugs, and combinations of drugs, are being developed to treat tuberculosis, as are new vaccines that may protect against disease in adults.  Quantifying the burden of TB is fundamental to understanding its global epidemiology and for making appropriate resource allocation decisions. Most estimates of new TB case numbers each year rely strongly on the number of cases reported by countries in that year to WHO. Unfortunately, one in three TB cases are thought to go either undetected or unreported, so the number of cases reported underestimates the number of new cases. While one can correct for this, it is hard to know exactly how much to adjust the reported numbers. Some countries have good systems for recording causes of deaths, which can be used to estimate the number of deaths caused by TB. Increasingly, large and expensive prevalence surveys are being used to estimate the number of people with active disease in a population. These estimates are less subject to bias, but measure a different quantity. Little work has explored the best way of combining these three data sources.  A major goal of this work is to use mathematical transmission models for burden estimation and provide a unified framework for all data. These models yield the number of new cases, deaths, and also the prevalence of disease. They explicitly represent disease transmission and so introduce a dependence between the number of new cases in different years. These models involve parameters evidenced from previous epidemiological work, but must be calibrated to learn from data on TB reports, deaths and prevalence. Calibration means adjusting imperfectly known model parameters in order to match observed model outputs to the data. This process provides a model that may be used to make predictions about burden, but may also teach us something about the underlying processes. Many of the parameters concerning the epidemiology and disease course of TB are quite uncertain, and this uncertainty is rarely represented fully in models needing calibration, but will be done in this project using statistical techniques that also allow comparison of different models' performance.   TB burden estimation and calibration of transmission models are almost always carried out on a country-by-country basis. Many parameters describing disease progression are likely to be similar in different countries, even if their exact values differ for unknown reasons. Hierarchical modelling techniques allow such parameters to be correlated between countries. This can improve precision, particularly for countries with little data, as estimates can be informed by data from neighbouring countries. I will explore these techniques for the transmission model, and also in statistical modelling aiming to account for the observed patterns of drug-resistance. The transmission model will ultimately be extended to include different types of drug resistance.  As new treatments and vaccines emerge, those with responsibility for public health will want to understand the potential impact these new technologies can have in terms of gains in health, and changes in spending. Producing cost-effectiveness and budget impact evidence requires a model that includes transmission, in order to account for indirect benefits accrued by avoiding secondary cases. We will use our model to provide guidance to decision-makers seeking to maximise health gain with limited resources.       "description_narrative":["A recent clinical study in Africa and South Asia has found that cryptosporidiosis is one of the most significant causes of death and illness from diarrheal diseases amongst children in the developing world. Cryptosporidiosis is caused by a single-celled protozoan parasite; the predominant species infecting humans are called Cryptosporidium hominis and Cryptosporidium parvum. This parasite mainly lives in the cells in the gut wall and has a complex life-cycle. Infection occurs due to consumption of water or food contaminated with the parasites. Parasites are spread from an infected individual through their faeces. In people who are healthy and well nourished, the disease clears naturally within a couple of weeks. However, in people who are malnourished (particularly in young children) and people with an immune system that is not functioning properly (for example HIV/AIDS victims), the disease can have a much more significant impact. It is the major contributor to life-threatening diarrhea in young children, with 2.9-4.7 million cases in children under 24 months in sub-Saharan Africa and the Indian sub-continent, leading to more than 200,000 deaths per year. Cryptosporidiosis is also associated with malnutrition and stunted growth in children and probably causes chronic infections, which last for weeks or months. The only drug registered for the treatment of this disease is nitazoxanide, which is not very effective, especially in those patients who are most severely affected due to a weak immune system and/ or malnutrition.   Therefore there is an urgent need for the development of new drugs to treat: (1) children <24 months, especially those that are malnourished and with chronic diarrhea; and (2) immunocompromised children and adults with advanced AIDS and chronic diarrhea. Cryptosporidium may be the cause of as much as 75% of chronic diarrhea in this patient cohort.   We have discovered some chemical starting points that can be used for a drug discovery programme. We have a series of compounds that kill the parasites and also are very effective in clearing the parasites from rodent models of cryptosporidiosis. The compounds are thought to work through preventing the parasite making proteins. The aim of this project is to take these starting points and optimise them to make a molecule which has the potential to be a drug. This will require us to optimise multiple features of the molecule: its ability to kill the parasite, its ability to reach the sites in the body where the parasite resides without being broken down, and its safety. At the end of this project we hope to have a \"preclinical candidate\". This is a compound that we think should be suitable to enter human clinical trials.       "description_narrative":["The continuing threat from new infectious diseases has been highlighted in recent years by the emergence of MERS-coronavirus in the Middle East, Ebola in West Africa and Zika in Latin America. Preparing for and responding to such threats is a priority for public health agencies and governments worldwide. Equally importantly, as scientific research delivers new approaches to controlling existing infectious diseases such as malaria, HIV, tuberculosis and polio, there is a need to determine what interventions are likely to work best to limit the disease and illness caused by a specific disease in a particular setting.  In this context, computer modelling of the spread of diseases in populations coupled with scientifically rigorous data analysis have proven themselves as powerful tools for giving insight into how diseases spread and what can be done to limit that control that spread. The Centre for Global Infectious Disease Analysis undertakes research on these scientific methods and applies them to disease threats of concern.   A key feature of the Centre is the close links it has made with with public health agencies - such as the Public Health England, the Centers for Disease Control and Prevention in the US (CDC), and the World Health Organization (WHO) - organisations which have to formulate and implement strategies to control infectious diseases.   Senior scientists in the Centre are world-leaders in infectious disease modelling and analysis. They have broad experience in responding to real epidemics, and undertaking epidemiological research on endemic diseases (such as malaria, HIV, tuberculosis and polio). Their work directly influences disease control policy through the many interactions the Centre has with public health organisations and governments.   Funding provided by MRC and Imperial College has allowed dedicated investment into the development of close collaborative partnerships with PHE, CDC and WHO and governments and research centres in many low and middle income countries (LMICs). We also work closely with other important organisations who work on improving health in LMICs (notably the Bill and Melinda Gates Foundation, The Global Fund and Gavi). With each, our collaborative research has both scientific interest to Centre researchers and contributes to the evidence base for public health decisions. Centre funding supports a core technical capacity available to all projects and pays for dedicated liaison staff who facilitate and enhance our partnerships with major global health organisations and LMICs.   MRC funding has also facilitated a greater focus on collaboration, training and career development within the Centre - training the next generation of quantitative infectious disease epidemiologists. We are also involved in increasing knowledge of advanced modelling methods in public health institutions and the wider infectious disease research community, and so have run a large number of introductory short-courses and workshops (in 13 countries in the last 5 years)  This applied activity depends on more fundamental research undertaken in the Centre on the transmission dynamics of different diseases, how diseases evolve genetically over time, and on analysing new approaches to control or treat diseases. This research is interdisciplinary, with Centre researchers working with leading clinical and experimental scientists around the world.  Continued Centre funding will allow existing partnerships to be sustained and enhanced, and new collaborations with researchers and health agencies in LMICs to be developed. By supporting PhD training and postdoctoral career development, it will also continue to expand the UK skill base in mathematical modelling and advanced analysis applied to health problems.       "description_narrative":["We seek funding to support an African Space Weather Workshop aimed at developing an international team to enhance capacity in space weather monitoring in low income countries.  The proposal seeks to utilise the world-leading capability of the UK in the field of space weather monitoring to investigate and address a specific issue impacting on countries on the DAC list. Space weather is a term used to describe the how solar-driven variations in the Earth's magnetosphere-ionosphere system can impact modern technology and thus human activity. To date space weather has primarily been studied over the polar regions. However, new emerging economies necessitates the elucidation of space weather in the equatorial regions. The most serious space weather threat to modern technological infrastructure is posed by geomagnetically induced currents (GICs) at the Earth's surface. GIC are driven by solar-wind/magnetosphere interactions which driven varying electric and magnetic field in space which again induce currents on the ground. Particularly powerful solar storms have resulted in power and communication failure. GIC can also result in currents being induced in oil and gas pipelines, leading to their degradation, of particular concern to oil-producing nations like Sierra Leone and Nigeria.  Further, changes in space weather will affect radio communication, as varying ionospheric conditions result in scintillation of radio signals. This can result in GPS and HF radio signals being absorbed, and significant GPS position and timing error. A particular challenge in the equatorial regions are high-density equatorial plasma bubbles which may span the horizon during severe space weather events.   Unique to equatorial region there is an ionospheric current system called the equatorial electrojet which may amplify space weather effects.   Disruption of radio communication will adversely affect both everyday life, commerce and disaster relief in equatorial regions. The University of Leicester are world-leading experts in space weather monitoring, using both ground-based and space-based instruments to probe space weather in the polar regions.  The research they produce is of an international excellent standard.  In this proposal we aim to utilise the University of Leicester's expertise in this area to bring together an international team of scientists and engineers for a one week African Space Weather Workshop at the University of Leicester.  The aim of the workshop is to form an international team focussed on increasing capacity for space weather monitoring, instrumentation, education and research in Africa.  The workshop will enable international networking, communication and knowledge exchange.  The workshop will identify the key challenges associated with African Space Weather Monitoring and develop a road-map for further collaborations.  The workshop will discuss how adverse space weather affects the African equatorial region, funding requirements, instrument development and deployment, and the steps needed mitigate serious space weather situation over the African equatorial region.  There will be a strong focus on identifying the training and education needs of scientists and engineers in the physics and engineering requirements associated with space weather monitoring.  This could involve planning and seeking funding for joint PhD projects between African and UK universities, academic exchanges, lecture tours of African and UK scientists, identifying instrument requirements associated with the monitoring of space weather, and engaging with local populations in building and deploying such instruments.       "description_narrative":["The overarching aim of ACREW is to develop the UK's capacity for building the resilience of developing countries to atmospheric hazards. The proposed programme of work will focus on two hazards:  air pollution and adverse weather - both of which disproportionately impact poor people.  Air pollution is a critical problem in many developing economies, reducing life expectancy by up to a decade, and GDP by up to 5% per year.   Weak infrastructure and dependence on rain fed agriculture render the populations of developing countries critically vulnerable to variation in the weather.    Recent technological and scientific developments offer opportunities to build the resilience of developing countries to hazards related both to air quality and to climate/weather.  In the case of air quality, new sensor technology promises a revolution in our ability to measure pollution on all scales, at low cost.  Reduced computing costs and readily available open-source software, furthermore, offer opportunities for scientists in developing countries to run their own air quality simulations.  In the case of climate and weather, newly available models, observational datasets and analysis tools have the potential to improve early warning of weather-related hazard on time-scales of days to seasons.   Users must, however, understand how much confidence can be placed in state-of-the-art models and technologies, if such systems are to be exploited safely in operational settings. This relies on sound scientific understanding of underpinning mechanisms, as well as on close engagement between researchers and end users.    All of the proposed work in ACREW contributes to the promotion of economic development and improved human welfare and health in developing countries. The specific activities have been chosen to exploit new scientific developments and existing expertise within NCAS. The air quality work packages (WPs) focus on the development of a new, affordable sensor for monitoring of air quality; liquid fuel composition and its effect on air pollution; and the formulation of evidence-based emissions policies through simulation of air quality in major cities.  The climate/weather WPs focus on early warning of agricultural drought; the risks associated with tropical cyclones; prediction of intense rainfall; and the utility of forecast information for managing complex systems, such as renewable energy supply and demand.  Each WP includes elements of science and application, although the balance varies depending on the activity.  Both the air quality and climate/weather components of ACREW include operational pilots of new technology, which will be carried out in partnership with on-the-ground organizations.  Strengthening and building of partnerships are key objectives for ACREW, and crucial for the project's success.  To this end, ACREW is centered round a series of workshops and exchange visits.  These engagement activities will cement collaborations and facilitate co-development of new technologies and research applications.  The participation of international organizations, such as the WMO, WHO, ECMWF and the Met Office in annual workshops will enhance the uptake ACREW scientific findings - widening impact beyond immediate project partnerships and pilots, and securing long-term development outcomes.","The Global Challenges Research Fund (GCRF) supports cutting-edge research to address challenges faced by developing countries.
