      "description_narrative":["\"This project seeks to assess how local communities and state authorities relate to the criminal groups that currently participate in the production and trafficking of hashish in Morocco and Spain. While it is well known that rebel groups and separatist movements provide goods and services as a means through which to contest state authority, it is increasingly evident that criminal groups with narrower objectives also govern people and spaces. Nevertheless, these groups rarely challenge the state directly. Instead, they tend to flourish in their interstices, seeking economic and sub-territorial dominion in order to make their economic activities both possible and profitable. Drug-trafficking organisations in particular tend to operate in places where states are seen to be weak or failing. Border regions and marginalised provinces such as the Rif in Morocco and Andalucia in Spain, both of which exhibit amongst the highest levels of unemployment and poverty in their respective countries, have been historically prone to hosting illegal activities (Pack 2019). Spurned by central government policies and incentivised by lucrative neighbouring markets, local communities have often accepted or supported the criminal groups that have turned parts of these regions into hubs for the traffic of hashish (Robins 2016). Who holds the monopoly of legitimate violence in these areas? How do social values and political circumstances shape local perceptions of the drug trade? Through what practices do criminal groups bolster support for their activities? These are the main questions this project seeks to answer.\n\nIt will do so through a host of qualitative methods including archival work, interviews and participant observation. Data collection will be focused, in each country, on the perspectives of local residents and their interactions with criminal groups and government organizations (see Case for Support). It will draw on the PI's experience conducting research and long-term ethnographic work in the Middle East.\n\nThe findings of this research will inform a broad academic audience in sociology, political science, geography and history. In scholarly terms, the project will make a major contribution to theories of sovereignty and illicitness, providing new insights into on-going debates regarding the links between corruption, social values and criminal governance. This will be achieved through an examination of how local communities in the Rif and Andalucia region relate to criminal groups involved in production and trafficking of hashish. Academic contributions will be delivered through scholarly articles, conference presentations, and a monograph. By encouraging a dialogue about the nature of drug trafficking and its links to local communities in the Rif and Andalucia, the project also seeks to improve development and crime prevention policies in these two countries. This will be achieved through a) research design and knowledge exchange workshops in Spain and Morocco b) op-eds in major media outlets in several languages c) a best practices guide that can be used by government agencies and policing bodies in the future.\n\nThe potential applications of the project stretch far beyond the two countries under study.       "description_narrative":["The programme will support low and lower-middle income countries to grow their economies in ways that help to protect and restore their natural capital and so drive sustainable economic development. It is designed to provide practical support to governments, businesses, and financial institutions to integrate nature into their economic and financial decision-making, understand and manage nature-related risks, and capitalise on growing opportunities to invest in their natural assets. As such, it will support low and lower-middle income countries to transition to nature positive, net zero economies and so protect the poorest communities. Through an integrated set of activities, the programme will deliver the following outcomes: • Private Sector Disclosure Readiness: private sector actors in low and lower-middle income countries – including financial institutions, businesses, and policy-makers - will have the tools they need to understand and manage nature-related financial risk. In particular, the programme will ensure that key institutions have the tools and capacity to respond to growing demand to disclose nature-related financial risk. • Integrating nature at country level: governmental and regulatory decision-makers in low and lower-middle income countries will have the knowledge, skills and data to design and implement policies and programmes that will help to manage nature-related risks, unlock new nature markets, and rebuild natural capital. • Action Plans for Nature: partner governments will develop clear and comprehensive plans to finance the protection and restoration of nature. These plans will act as platforms to mobilise and guide both public and private financial flows. • Evidence Sharing Mechanisms on Nature: better evidence will be available to, and used by, decision makers in low and lower middle-income countries to guide their work. The programme will help to build the evidence about how to best integrate consideration of the natural environment into economic and financial decision making. It will also help decision-makers in governments and the private sector to access and use that evidence easily by building communities of practice and robust approaches to sharing knowledge and information. The outcomes will support the implementation of the Kunming-Montreal Global Biodiversity Framework (KMGBF), agreed at the Convention on Biological Diversity (CBD) meeting COP15. As protection and restoration of critical ecosystems is also critical to tackling climate change, it will also support the UK goal to keep global temperature rises within 1.5c degrees."],

      "description_narrative":["Neglected tropical diseases are a diverse group of diseases that thrive mainly among the poorest of the world's populations. Three of the World Health Organisation's 10 most significant neglected tropical diseases - African trypanosomiasis (also known as sleeping sickness), leishmaniasis, and Chagas' disease - are caused by closely-related single-celled parasites. Human infection with African trypanosomes is brought about when an infected tsetse fly takes a blood meal. Without treatment the disease progresses through general ill health to coma and death. Current drugs in use against trypanosomes are old, toxic and failing due to emergence of resistance. Urgent new research is needed to identify potential new therapeutic options.  An unusual aspect of African trypanosomes is that they multiply in the human blood in full view of the body's defence systems. They do this by periodically changing their cell surface to escape recognition by the host. However, many molecules on the parasite surface perform essential functions and cannot be changed. Trypanosomes place these in a special, protected domain on the cell surface. Identifying surface-exposed invariant molecules and understanding how this protective segregation is maintained are of major scientific interest, as well as of practical utility in uncovering ways in which trypanosomes may be vulnerable to new therapies.  Using a combinatorial approach funded by the MRC, I have previously identified the composition of the African trypanosome cell surface. I now aim to exploit this knowledge to single out those invariant surface molecules that are essential to the survival of the parasite during infection. For this, I propose to harness some of the power of modern DNA sequencing technologies to test which cell surface genes, when silenced, cause the parasite to die inside the host. A similar method will be used to identify not only cell surface genes, but any genes that the parasite uses to maintain the cell surface organisation, which is so critical to escaping the host immune attack.   Finally, I propose to test those surface-exposed molecules for their potential as vaccines. My pilot experiments show that I can use genetically-modified parasite to screen for molecules on the parasite surface that are accessible to the immune system. I propose to develop this assay and test the most promising candidates in an animal model of human disease.       "description_narrative":["The proposed research concerns the most important human malaria parasite, Plasmodium falciparum. Malaria is one of the world's most debilitating infectious diseases, killing over half a million people every year and affecting several hundred million. Most of the deaths occur in young children in sub-Saharan Africa, but adults can also suffer from malaria throughout their lives, reducing quality of life and retarding economic development in endemic countries. The lack of an effective vaccine and the emergence of drug-resistant parasites mean that there is now an urgent need for research leading to a better understanding of the malaria parasite, and hence to new vaccine targets and treatment strategies for this disease.  The malaria parasite causes illness via the infection of red blood cells. It multiplies inside these cells and modifies their surfaces with proteins called PfEMP1s that bind to the walls of blood vessels. This is crucial for parasite survival as it removes infected cells from the circulating blood and protects them from passing through the spleen, which might recognize and destroy them. It also contributes to disease, with severe malaria being particularly associated with the accumulation of infected cells in vessels of the brain and placenta. It is therefore of great interest to malaria biologists to understand the mechanisms that control the expression of these adhesive PfEMP1 proteins. PfEMP1s are not expressed uniformly by all malaria parasites: instead, individual parasites regularly switch between different variants. This allows them to stay ahead of the immune system and sustain a chronic infection for months or even years. The parasites have a large, variable family of 'var' genes for different PfEMP1 proteins and they vary the expression of these genes by so-called 'epigenetic switching'. Furthermore, var genes recombine very readily to generate new variants, so each parasite strain - of which there are many hundreds circulating in endemic areas - has a unique repertoire of possible surface proteins. This is one reason why immunity to repeated malaria infections is slow to develop in humans: every new parasite strain looks different to the immune system, so people can be re-infected repeatedly throughout their lives.  Understanding and ultimately interfering with the expression, switching and recombination of var genes, and thus the variant expression of PfEMP1 proteins, could be a key to more effective immune control of malaria. Therefore, this research focuses on a new biological mechanism that the parasite may use for switching between var genes and for generating new variants. Our recent work has showed that an unusual DNA structure called a G-quadruplex that is concentrated around var genes seems to affect both these processes. To investigate this further, we now propose to map the G-quadruplexes throughout the Plasmodium genome, both in DNA and also in the messenger molecule, RNA. We will use a range of cutting-edge genome-wide technologies to do this, and will then check whether the distribution of the structures changes when the enzymes that unwind them are removed.  These studies will lead to a better understanding of the mechanisms underlying var gene dynamics, and may ultimately inform new strategies to combat malaria, since var genes - and the adhesive proteins that they encode - are central to malarial disease. The outcomes of the research will be published in open-access scientific journals and presented at international conferences. They will be communicated to the general public via summaries on appropriate websites and via science writing in magazines and/or online.       "description_narrative":["Tuberculosis (TB) is a major cause of disease and death globally. In 2015, WHO estimated there were 9.6 million TB cases and 1.5 TB deaths. Nearly 500,000 of these cases were resistant to two or more of the main drugs used to treat TB. New drugs, and combinations of drugs, are being developed to treat tuberculosis, as are new vaccines that may protect against disease in adults.  Quantifying the burden of TB is fundamental to understanding its global epidemiology and for making appropriate resource allocation decisions. Most estimates of new TB case numbers each year rely strongly on the number of cases reported by countries in that year to WHO. Unfortunately, one in three TB cases are thought to go either undetected or unreported, so the number of cases reported underestimates the number of new cases. While one can correct for this, it is hard to know exactly how much to adjust the reported numbers. Some countries have good systems for recording causes of deaths, which can be used to estimate the number of deaths caused by TB. Increasingly, large and expensive prevalence surveys are being used to estimate the number of people with active disease in a population. These estimates are less subject to bias, but measure a different quantity. Little work has explored the best way of combining these three data sources.  A major goal of this work is to use mathematical transmission models for burden estimation and provide a unified framework for all data. These models yield the number of new cases, deaths, and also the prevalence of disease. They explicitly represent disease transmission and so introduce a dependence between the number of new cases in different years. These models involve parameters evidenced from previous epidemiological work, but must be calibrated to learn from data on TB reports, deaths and prevalence. Calibration means adjusting imperfectly known model parameters in order to match observed model outputs to the data. This process provides a model that may be used to make predictions about burden, but may also teach us something about the underlying processes. Many of the parameters concerning the epidemiology and disease course of TB are quite uncertain, and this uncertainty is rarely represented fully in models needing calibration, but will be done in this project using statistical techniques that also allow comparison of different models' performance.   TB burden estimation and calibration of transmission models are almost always carried out on a country-by-country basis. Many parameters describing disease progression are likely to be similar in different countries, even if their exact values differ for unknown reasons. Hierarchical modelling techniques allow such parameters to be correlated between countries. This can improve precision, particularly for countries with little data, as estimates can be informed by data from neighbouring countries. I will explore these techniques for the transmission model, and also in statistical modelling aiming to account for the observed patterns of drug-resistance. The transmission model will ultimately be extended to include different types of drug resistance.  As new treatments and vaccines emerge, those with responsibility for public health will want to understand the potential impact these new technologies can have in terms of gains in health, and changes in spending. Producing cost-effectiveness and budget impact evidence requires a model that includes transmission, in order to account for indirect benefits accrued by avoiding secondary cases. We will use our model to provide guidance to decision-makers seeking to maximise health gain with limited resources.
