Consequently, the effects of long electricity blackouts have demonstrated impacts on economic activities and social stability and security. A framework for disaster management and resilience of the power sector is needed, beyond the occurrence of \"average\" outages contemplated in current security standards. This framework should consider network management under the occurrence of natural hazards such as earthquakes and tsunamis that may cause major blackouts, and assess proper measures to manage the associated disasters.       "description_narrative":["The mucosal surfaces (skin, intestine and gills) of fish are constantly interacting with a wide range of pathogens and, in concert with the resident mucosal microbiota, are the first line of host defence. Several immune factors have been described in fish mucus and their presence and abundance can be affected by pathogen infection. As a consequence, the mucosa-associated microbiota can be affected and this may lead to dysbiosis, a microbial imbalance on or inside the body, which reduces the beneficial role of microbiota on the innate immune system and may lead to secondary infections by opportunistic microbes. Sea lice (Lepeophtheirus salmonis and Caligus spp.) infections are the most economically important causative agents of disease in marine-farmed salmonids worldwide. Caligus rogercresseyi is the main ectoparasite affecting the Chilean salmon industry with estimated costs of up to $120 million annually. This parasite damages external surfaces of the integument providing portal of entry for secondary infectious pathogens, and infected fish also become stressed which leads to immunosuppression with consequent increased susceptibility to the secondary infections. Other Caligid parasites (Caligus rogercresseyi and other Caligus spp.) have detrimental impacts on mariculture  operations in both developing and developed parts of the world which causes economic losses of US$ 100's millions annually. The development of novel methods to prevent or reduce sea lice infection is therefore a high priority for the industry. An essential prerequisite for improved treatments strategies is a better understanding of relationship between host-microbiome-parasite at the epidermal interface. Therefore, the aim of this research programme is to evaluate the influence of sea lice parasitism on mucosal health, including aspects of the immune response and host microbiota. We will evaluate the effect of immunomodulants on susceptibility to sea lice infection, and on gut and mucus quality by determining epidermal mucus proteomic responses, microbiota changes and mucosal transcriptomic changes.       "description_narrative":["The economic burden of infectious disease hinders sustainable farmed Atlantic salmon production in Chile, and most aquaculture species worldwide. Targeting production of resistant stocks by selective breeding is a long term goal, and will contribute to disease control at a population level. Enabled by advanced genomics tools and technology, genetic improvement of disease resistance can be accelerated using genetic marker data to identify resistant parent fish in breeding programs. Further, the large families and the practical feasibility of large scale disease challenge experiments make salmon excellent models to discover genes and mutations underpinning host response to pathogens. Focusing on two of Chile's most problematic diseases (sea lice and Salmon Rickettsial Syndrome; SRS) in Atlantic salmon, this project will harness these genomic tools to (i) discover genes and functional variants affecting host response to infectious disease in farmed salmon and (ii) improve the use of genomic tools in breeding programs to increase population-level disease resistance via genomic prediction, and reduce the negative impact of outbreaks.  There is now a substantial genomic toolbox now available for Atlantic salmon (e.g. an advanced reference genome assembly and high density genetic marker arrays), this offers an unprecedented resource to locate specific genes and causative genomic variation underpinning host response to pathogens. In the proposed project, we will utilise data and samples previously collected from two large-scale disease challenge experiments (sea lice and SRS challenges. Genome-wide genetic marker data will be generated for all animals, and locating functional variants for resistance to each of the diseases will be achieved using the complementary approaches of (i) high power genome-wide association analysis; (ii) RNA-Seq (gene expression) comparison of the most resistant (R) and susceptible (S) individuals; (iii) Whole genome resequencing of pools of the R and pools of the S individuals combined with functional annotation. In addition to improving knowledge of the fundamental biology of host response to pathogens, these results have potential commercial relevance in at least two important fields. Firstly, the genes identified and their encoded proteins are potential drug or vaccination targets. Secondly, genomic regions and specific variants can be incorporated into selective breeding schemes, via improvement of the accuracy and cost-efficiency of genomic prediction of disease resistance.       "description_narrative":["This project involves collaboration between two teams of experts at the University of Aberdeen and University of Santiago of Chile, conducting state-of-the-art research in complementary areas of fish immunology/fish vaccination.  Aquaculture is one of the fastest growing sectors that provide food to the expanding world population. It is estimated that ~50% of fish consumed worldwide are farmed, and this is projected to rise. Sustainability of fish farming relies on good management of fish health and control of diseases. Vaccination is an effective strategy to control many common diseases and many highly efficacious fish vaccines exist. However, the development of fish vaccines has been largely empirical, based on whether a formulation is effective at increasing survival post-disease challenge. This is unsatisfactory from both ethical and scientific perspectives. There is a clear need to establish methods to improve fish vaccine development.  This project will undertake studies to characterise an important immune pathway that may be of importance for future fish vaccine development. Vaccination relies on the stimulation of adaptive immunity in vertebrates, with long-term memory responses giving protection when encounter with a pathogen occurs. In mammals a key effector population driving such responses are T helper (Th) cells, that release intercellular mediators (cytokines), that initiate antimicrobial responses, including antibody production. These responses have to be tailored to the pathogen type, with viruses, parasites and extracellular bacteria requiring different immune mechanisms to give protection. Different Th subpopulations differentiate in response to these different pathogen types and host factors, and release different repertoires of cytokines to produce the most appropriate response. We know virtually nothing about these responses in fish, although many of the genes involved in mammals are now characterised or have putative homologues likely to have equivalent function. For example Th cells express CD4 on their surface, and two types of this molecule exist in teleost fish, that will likely define these cells. Here we propose to develop antibodies to the two CD4 molecules to detect, isolate and characterise the CD4 subsets (CD4- 1+, CD4-2+, CD4-1+/CD4-2+). In immunised fish, we will study their ability to express different cytokine repertoires upon restimulation in vitro with specific antigen. Sorted CD4 cell populations will be analysed. As antigen, both a bacterial and viral protein will be used in rainbow trout, an important farmed fish in both Chile and the UK. We have developed many reagents (eg primers for immune gene expression analysis) and immune proteins (eg recombinant cytokines) for use in trout, and expect the responses to be representative of those in salmonids more generally. Following the initial experiments, we will study the effect of adding different cytokines together with the specific antigen on the ensuing responses, by analysing cytokine gene expression and CD4 subset variations. We will next select the cytokines showing the most marked effects on directing these responses to link back to confirming the involvement of CD4 (putative Th) cells. This will be done by analysing cytokine gene expression in the sorted (CD4) cell subsets following antigen restimulation in vitro in the presence of recombinant cytokines. These results will go a long way towards confirming the function of Th cells in fish, and will establish if they express different cytokine repertoires in response to specific antigen and cytokines. We anticipate these responses will be of value as markers of protection in future vaccine development programmes, helping to improve the efficacy of poorly performing vaccines, and to generate vaccines to emerging diseases.
